I would put the sugar flowers on before serving, rather than overnight, esspecially if its going in the fridge, cause the moistre in the fridge and in the icing can dissolve the sugar flowers.
Icing the cake 4 hours before the party would be fine
Another option is to make the butter icing the night before, cover and put in the fridge. Get it out a few hours before icing and let it sit at room temp to soften, give it a beat and then ice, that way you don't have to make it all up before the party as well as ice it
I second putting the sugar flowers on at the last minute.
The colour (assuming its different) could bleed of they are on for too long and in the fridge.
