Meredith - I think you can't compare your body to a lab - I think the 2 inside you have a great chance of giving you a BFP and a H&H baby. At the end of day, a lab is a lab - they don't understand everything about a woman's body so how could the possibly replicate it no matter how good they are.

Ishy - I hope the BT gives you the definitive BFP that you deserve (please let us know - we'd be so excited for you). You had to make a horrible decision....gosh, the more I info I have from these posts the more I think that PGD is the way to go and if it means multiple cycles to get a good embryo - so be it...I don't want to miscarry again nor do I want to be faced with the same type of decision you had to make even knowing that we'd do the same...but it would be heartbreaking.

WTH: Cyclebuddy hope the DHEA does wonders for you. I was on Gonal F last cycle, the needle is really fine and didn't hurt at all.
AFM Already nervious about the next cycle. My FS said she'll start me on 300, wondering if she shouldn't go a bit more aggressive from the start, last time I was on 262.5 from the start and then went up to 375 without any effect. Anyway perhaps may have to insist on an early BT and then to get really more aggressive if E2 number are not up.

Sara - quick note - I think you should ask your FS about the dosage just so you understand why they are picking that level. I don't know much about dosage except for my own experience first flare cycle - started at 350 gonal f - after first follicle scan she pushed up to 450...2nd flare cycle - she started me at 450 ...3rd cycle which I hope to begin in Jan and will be antagonist, script again says 450. It may be that if you get good results from lower dosage, they don't want to give you unnecessary hormones. However, it was my understanding that they start low so that you don't get OHSS (or whatever the acronym is over having too many follicles) but that this is not really a risk with older women.
WTH - the gonal f needles didn't bother me at all - found it very easy to do - didn't even feel the needle, when the medicine goes in it burns a tiny bit, but nothing too bad. I even thought I'd want my husband to do it, but ended up finding it easy to do myself. did you get a dvd from the chemist? it's handy to watch before you do the first shot. if not, the little pamphlet that they give you is helpful (if you didn't get that, I'm sure if you google gonal f along with the name of the drug manufacturer their website will have a copy of the pamphlet).

Buliej - thanks for the tips on Gonal-F. Is it not just like the Puregon Pen? I guess what I am most interested in is whether it actually IS stronger than Puregon. My response to Puregon was reasonable. According to my FS, trying Gonal-F could be for better or worse. Have you heard if Gonal-F is better? I have been told they are both FSH, but there are "more sugars" in Gonal-F, whatever that all means....
Sara - are you switching to Puregon? I am REALLY surprised by how much your FS is holding back on your next cycle. My first Puregon cycle, I started on 200, was upped to 300. Cancelled, and next cycle put on 375. It is true that you can have too much FSH and sacrifice egg quality. But it does seem very conservative. Any chance of a second opinion? Do you feel confident at perhaps questioning this?
Ishy - sorry for your sad loss and what you have had to endure, but..... :dance: CONGRATULATIONS :dance: on your BFP :clap: Some of the ladies here have had to make similar difficult decisions in the past so they understand what you have been through. Just like you, that is what brought them to PGD. You are the first official BFP on this thread and give us all such hope. Wishing you a happy and healthy pregnancy :D

Sara, I hope FS is monitoring you closely so if the respond is not as desired she can adjust straight away. To my understanding, no Lucrin this cycle right, so that's an factor could make the difference from the last cycle.
Ishy, so glad you had your BFP after a hard journey, you totally deserved this! Enjoy the excitement and hope this is the first one in this thread and Meredith will be the next.
Hi to BW and Buliej, thanks for thinking of me.
I feel relieved that the worst part of a stim cycle is over, at least physically. EPU this morning went really well we were back home before 11am, I have been drugged up to take the edge of the pain and had been sleeping a lot until now. I did saw some blood during the procedure, had one glimpse of the tube of fluid from my follicles and quickly moved my eyes away from them...I prefer to ignore them than freaked out. No bleeding afterward which is good. Pain relief worked well and I feel quite pain-free at the moment.
We've got 9 eggs, 2 immature, so 7 really as I predicted. Embryologist did say that they will still put all of them including the 2 immature ones for fertilization in hope that they will grow in the mean time. So we will hear what's the fert report tomorrow morning.
It will be interesting to see this cycle how many blasties we ended up with comparing to last two cycle. Assuming it was DH's sperm DNA affected past results, now with the improvement in this parameter, we are hoping to see some difference. I truly hope that this is just that little tweak does the trick. Thanks for all the support, I really can't do it without you guys..now the real torture of the mind begins...hehe

Cuddlepie - 7 sounds great! Just curious - where you not under anesthesia during the pick up ... it's sounds like you were fully awake - I've always been knocked out (they say light anesthesia - but I'm completely gone).
WTH - I've never used Puregon, only Gonal F - so can't speak as to the difference...I'm not sure if there is a "stronger" - my guess is that people respond differently to different drugs even if they are mostly similar drugs...just another one of those IVF things that seem so variable
Looks like there will be a few of us starting afresh in January - hopefully it will be a lucky month...I do hope AF arrives for me in the next 2 weeks - if it doesn't that will really throw me for a loop.

You got me confused I always thought Gonal F is Puregon - but now checked in the fridge and it said Follitrophin alfa or similar.
WTH I think it might be a good idea for me to use the other drug instead since I didn't react at all on Gonal F - it's probably different from person to person. I thought there was nothing wrong with your reaction on the other drug in the second cycle. But I think in these things we may just have to trust our FS, I have decided to go with what she says for the start but to insist on an early BT/US if they don't plan that anyway and if my FSH is not up to scratch to question if my dosage shouldn't be increased straight away.
Buliej: My nurse told me if AF doesn't come as expected to come in for a BT as they would be able to tell me where I am up - if you're nervous about AF, maybe check if you could do a BT, it might give you some more security. I've decided to go shortly before Christmas or in between the Years just to see how I am going as my clinic will be closed on Christmas Day and New Years Day and we want to go to the mountains for two nights over New Years Eve, would be a real pain if AF decided to come on one of those days.
Cuddlepie: yes there won't be any Lucrin and I am not on the pill so should be start with my normal hormones which hopefully makes all the difference. Hope you're still relaxing and enjoy having DH around - he may have to make fruit juices for you ;)
Do you have a few days off (I thought you said that you did with your other cycles) or is it straight back to work on Monday?

Sara, puregon and gonal F are essentially the same drug, just made my two different companies. There may be slight differences because some people respond better to one than the other, but they essentially serve the same purpose.

Just a quick one, my ladies...
Fertilization report this morning showing that 6 of our eggs are fertilized. So out of 7 mature eggs, I think that's right on par with our fert stat (80%) :D the crunching time will be from Day3-5 though, fingers crossed. I am praying so hard that we will get more blasties than before....
Buliej, Im with SIVF, they do sedation first and then local anesthetic, so yes, I am awake the whole time, just feeling dizzy in the head like you are drunk, it was actually really good. I can look at the screen to see the needle going in to suck all the fluid out and then can see the eggs showing on the screen, its pretty cool. And doesn't take me long to recover afterwards. I think this is general practice in SIVF unless you ask for a general....
Sara, I am hoping to go back to work tomorrow. I was on painkillers yesterday but none today and feeling ok. I thinking going back to work just make time go faster IYKWIM...

cuddlepie - those numbers sound really good - fingers crossed that they keep going and get A+'s on their tests! I may ask about the light sedation - I hate the time they make you spend in recovery after anesthesia - I'd like to avoid that and also wouldn't mind seeing what is going on.
Butterfly Warrior - so exciting for you.
Good idea Sara - if no AF by say the 18th - I may ask for a test to make sure my pregnancy hormones are back to 0 as they should be.

CP - great job on getting 6 lovely embies. Praying you get at least half that in good blasties. I was right out for my EPU to but would find it really interesting to see what goes on.
Ishy - A BFP, thats fabulous news. Hoping you have a trouble free pregnancy!
Sara - I would say that your FS put you on a conservative dose because you are doing antagonist cycle so no supression at all. And as BW has said, Gonal F and Puregon are both exactly the same thing (FSH) just different makers. The general theory is that you should really respond to both just the same but it does seem different in certain individuals, Interestingly, my clinic told me that puregon is actually slightly "stronger" than gonalF. I was written up for 225iu of GonalF but since I'd used Puregon every other IUI cycle, they just changed to puregon but dropped dose to 200.
WTH - You might be interested in what I just wrote to sara about GonalF vs Puregon. Is just what my clinic told me but who knows? I really hope you have some "fun" over xmas and don't need your next PGD cycle
BW - wow, only 6 weeks to go. Counting down the days yet? I bet you can't wait to meet your precious little bundle
AFM - still feeling a bit disheartened by this whole thing although trying to have hope. I truly don't know what we will do if I don't fall pg. Not sure if DH will let us part with the $$$ for another PGD cycle but I am a touch scared about going back to "blind" OI/IUI's. Only 1 more week to go until BT. This part of the TWW really, really drags!

Meredith: thanks for the tips on Puregon - my FS doesnt' seem to give any instrutions to the nurses about which of them I am using, so may ask about Puregon next time, especially as a had a bad headache with the higher dosage of the Gonal F.
AFM: Probably still really innocent in this whole experience. Just really realised this weekend how stressful cycling is, really felt like my energy was coming back and that things that were in the too hard basket when I was jabbing are easy now.

Sara - I get you on the stress thing. It is really hard to functions normally when you are pumped full of hormones plus all the anxiety over what is or isn't going on. Ah would be the joy to be one of those women who just finds out they are pg at 12-16w and everything is all fine and dandy. hard to believe they exist but there are certainly plenty out there!
WTH - thanks for the cheer. Wish I just knew already!
Realistically, I am either pg or not by now so just want to move on either way.... sigh.... I am normally very patient but not with this stuff!
Can't say I have a strong feeling for yay or nay yet. Got all sorts of symptoms but with all the drugs, who knows?! Pessaries don't usually stop AF so will certainly know by Saturday when she's due.

Sorry for being a bit slack ATM, haven't been feeling very well yesterday. I was sure that I am ok to be at work given I didn't take any painkiller on Sunday and feeling alright, but it turns out to be the longest day ever. At the end of it, I had bad back pain and felt a bit heavy at lower abdominal area. Didn't have a good sleep and still in crap crap mood today. I am suspecting because of the sudden drop of the hormornes...This morning, got a faint brownish discharge (sorry TMI) and maybe the old blood from the procedure just came out? Back pain is still here and I will see how I can stand at work today.
I had a good old rant to DH last night and think I just can't deal with all these anymore...anyway, today is a new day. I will update you how we go on the embie report later.
Thanks Sara, Meredith and WTH for thining of me. How can I do it without you? Sometimes it just seems hard. Meredith, I hope you are holding on alright, at this stage, no news is good news I guess. When is b/t?

I went to the Christmas service on Friday night, it was really lovely and really sad. We were given a gift box that we wrote our loved ones name on and then hung it on their giant tree, and we took them home afterwards. It is now on our tree here. It is wonderful how strangers unite when they have something in common, I was sitting at the front on the end of the pew so everyone had to walk past me to hang their box on the tree, I was crying and I had 2 women touch my shoulder and offer me their sympathies, and another lady took my hand and smile. I cried lots that night but was really happy I went.

Clinic just called about my 6 fertilized embies. 2x7cells, 1x6cells, 1x5cells, 1x4cells. So total 5 are still vaiable and two are a bit slow. I hope they will catch up soon.
This is the time that I wish that my clinic will just do Day3 transfer. Then I will have more to freeze etc etc. I know from D3 to D5 will have a big drop, with my spirit at the moment, I will feel very lucky to have a good blastie to go in on Thursday.
Lisa, I am so glad to read that you enjoyed the service and it must be beautiful and I have tears in my eyes. Have a good cry and grief your angel and I hope you find it make it a bit easier during this silly season.

CP - those numbers sound good to me - as I'm new to the whole PGD thing - when will they test the embryos? isn't that why they can't do the day 3 transfer or can some clinics do PGD early enough so that you can still do a 3 day transfer?

Buliej, because this is our first cycle with my FS, he wants to see how many embies we ended up with for this cycle, so this cycle we won't be doing PGD. FS suggest it because of my age and past m/c, but only when we have sufficient embies....so obviously there is not enough and we can't risk with no embie go to TF. Tough tough tough....:rolleyes:
I think it is a general practice in SIVF to do 5 day embyros including PGD biopsy on Day 5. but I think in Melbourne, some clinics do Day3 biopsy which is what Meredith did....

Apparently my FS is swapping a lot of clients across at the moment. She says it is still pure FSH so I should respond similarly. I guess we will find out in a month's time... :pray: for you and your little beanies

Lisa - the service sounds very healing. It is lovely those women reached out to you (in only a way that women can). I am glad you are finding some much needed support in all corners. Seems very timely :)

AFM - surprisingly perky! Wasn't expecting to do so well after a failed IVF cycle! Perhaps it's knowing that I only have to wait one month until I start stims, rather than unnecessarily blowing one with the down-reg protocol. Have bought some fresh pineapple for DH to make his swimmers happy for proposed BD-ing this weekend :redface: Meanwhile, antibiotics kicked in right away so seem to have nipped the UTI in the bud. Was assured the drugs were Class A and would not interfere with my follies. Am on Day 8 now and usually have my estrogen dip bleed/cramps today or tomorrow. :pray: the MacaRoot/accupuncture/two months of stims do the trick and I make it through!

CP - big hugs to you atm. I can certainly understand your mood..... the things we put ourselves through eh? Sounds like your embies are on track for day3 but I know the next 2 days will be the real test. I am kind of with you on the whole day5 thing. I KNOW its meant to be gold standard and helps weed out the best embies, gives the highest pg rates etc etc but I just don't believe that every emby that didn' t make it to day5 would not have made it full stop. My sister did IVF 12 yrs and 5yrs ago ( that one with donor eggs) and her ET were all day 3. Neither of her now healthy children were good looking embies at all ( even at day3) so you womder if they would ever have made it if left to grow on. I guess I am coming now from a viewpoint where none of my 9 "good" embies made it to quality enough blasts to freeze so the whole "drop off" is really, really scary.
Praying you get at least 2 good ones so you have a spare ( or another sibling) on standby!

Me - not much to report. Days dragging on and on as I count this last week down. DH is never that fussed by my symptoms etc after a cycle but this one has really got to him and he told me last night how desperae he was for this to work. We still haven't really discussed what to do if we get BFN. I think he likes the idea of doing PGD again but the costs understandably concern him.
I think I am really feeling the "pressure" of needing to get pg this time to get our PGD tested baby.

Good morning, girls. Could not get on BB at all yesterday afternoon so I guess that's why there's been no action here for a record 2 days!

Meredith - yep, that symptom analysing will definitely do your head in. Are you thinking of doing a HPT around the time of the BT? I just could not handle hearing it over the phone so chose to do one with DH when I knew the Pregnyl was well and truly out of my system but a test work work (15 dpo). I wanted to share the moment with DH - whether the outcome was joyous or sad. Just curious...
AFM - well, my spotting and cramping arrived right on cue, at midnight between Day 8 and 9m as it has since all non-PG cycles since Jan 2008. Quite light, and continued until after breakkie. Then returned for an hour again last night. But it has been heavier in the past on cycles where I have naturally fallen PG. I did spot for a couple of hours last month on Day 7 FSH, despite the fact my estrogen was well over 600. So maybe no harm done to my follies this month. Maybe the maca root and accupuncture have helped a little bit. I have spoken to two FS about this and neither will help or comment. They just say it won't happen on IVF (even though it does). My theory now is that around Day 8 or 9 the dominant follicle is trying to take over and perhaps the other follies stop producing as much E2. Only this does not explain why it still happens on an IVF cycle. Alternatively, maybe it is light because it has taken a few days for the lining to build only to be shed again.The more I think about it, the more it does my head in... :wall: End of my long-winded rant. I just don not know if this happening to anyone else and just wish my body would act more normally and continue to fear this is all pre-menopausal.

Sorry not really up to personals today althoug I am hoping CP has had a sucessful transfer today.
Past 2 days I had beun to feel fairly confident i was actually pg. Usually i would have very strong AF cramps by now - not happening, plus I've had sore BBs and yesterday felt quite nauseus at times.
With all my other IUI cycles, this has always been a great sign and I have always done HPT around this time (12dpo). My "gut" feeling has always been right and if I think I am pg then have always been greeted with two strong lines.
Well, feeling confident, I did HPT today. NO second line. Not even a hint of a second line when I squinted!.
So I would have to say a defnite BFN for us and must be the whole IVF which is messing with my body. I know everyone will come in here and say "its to early" or "don't give up until your BT'" but i KNOW there is no chance. I have been pg 5 times and even though all but 1 were not sucessful, they all gave good strong BFP on HPT so a -ve HPT IS a BFN for me.
I am more shattered than i ever expeced to be. Had to call DH ome from work. I was sooooo hopeful for this cycle and just still in shock I guess. This year has just been bad news after bad news after bad news. I just don't know how much more I can take.
To add to that, I am stressing already about what we do next. DH thinks we should just go back to IUI and take our chances without PGD. He is worried not only b the costs, but also thinks all our embies were damaged by the testing. I can see his point given our "drop off" from 14embies and none frozen but I am also terrified of having more m/cs. Haven't got appt with FS until the end of Jan. I REALLY need some questions answered before then.
So sorry for the rant, I know everyone in here has faced failed cycles. I think I am just reaching the end of my tether at seeing our dreams slipping away faster by the day.

Still hoping that there is still a chance that the test is wrong but can understand how devasting it is.
Perhaps you could ring your FS and ask if they could slot you in earlier in case they have a cancellation or if you at least could talk to your FS over the phone. After my cancelled circle I was able to talk to the nurse about my options which also was of some help.
My FS also sometimes answer my questions via email, so maybe you could ask if that would be possible.

Meredith, I am sorry to hear your news but really hope you are wrong. I know you don't want to hear this but until you have b/t please don't lose hope. I can totally understand 'reach the end of tether part', I was so feeling the same as you yesterday and had a big full on cry. I just feel I can't face this anymore...
Anyway, that cry must have helped me to face the TF day, I am feeling better. TF went well although we were an hour late due to so many scheduled in today at clinic.
Out of my 6 viable embies on Day 3, one is a front runner, a grade 2 blastcyst, 2 early blasties, the rest 3 are still in Day 3 cells-stage they said but they will keep an eye on them to see any more development. I am relieved about the result to say the least. Also happy to see improvement in the result that seems DH's improved SCSA result did make a difference from DAy3 to Day 5 development, normally we would see a huge drop off and most of them are fragmented.
FS suggested to transfer two, the grade 2 blastcyst and 1 early blasties, he said because we don't know whether in the next two days they will definitely be frozen, they are still perfect for transfer at this stage. The chance of multiple pregnancy is quite low and we don't want to waste a perfect embryo. So here I am with 2 on board now. We've never done 2 before I know it won't increase my chances but I am still excited. Hope they will grab on and stay for a long haul.

Cuddlepie - I'm very excited for you...Also, I think 2 does increase your chances - my FS originally suggested transferring 2 to me...and we did the first time when we had 2 to transfer...for my next cycle if we don't do PGD I probably won't do 2 but if we do get to do PGD and end up with 2 good ones I most definitely would ask for both of them to be put in.
